I wonder if this is meant to be a real place or not?

Take your pick, I guess...
Hazleton North
Hazleton South
Or maybe one of the Leygore Manor tombs?

Someone at the Society of Antiquaries of London says it's probably Notgrove...
"The plan of the chambered tomb which Indiana has chalked up on the blackboard as Turkdean appears to be based on (but not an accurate representation of) that of another Cotswold tomb, the Notgrove long cairn excavated by Elsie Clifford in 1934–5 and published in Archaeologia for 1936 (which is the year in which the action of the film opens – and, if I am not mistaken, Indiana has a copy of Archaeologia on the desk in front of him while he is giving the lecture!)." - SALON.

Notgrove is not known to have been 'one of a pair' but the others Hazleton North/South and Leygore Manor 1 & 2 are each thought to be 'pairs' (and thus worthy of scholarly discussion) -

But play the film for about 30sec longer - the reference to folklore and The Golden Coffin is taken from the legend that relates to a Round Barrow in Far Oakridge (mentioned as The Golden Coffin on this site MegPort) -

Notgrove Hazleton and Turkdean are adjacent parishes but The Golden Coffin is in Bisley (with Lyppiatt) some 15/20 miles to the West -

Now could the Lost Ark be somewhere in Gloucestershire?

So, it's a hodgepodge of real places. The diagram is a truncated version of Notgrove and the legend is from Far Oakridge.
